Altered DNA methylation has been reported in cancer cells and may be a critical event that drives a variety of cancer processes. The aim of the study was to screen for serum methylated proteins in patients with metastatic anaplastic lymphoma kinase (ALK) inhibitor-resistant non-small cell lung cancer (NSCLC). Protein expression profiles in paired tissue and serum samples of patients with metastatic ALK inhibitor-sensitive (n = 18) or -resistant (n = 7) NSCLC were analyzed by 2-dimensional difference gel electrophoresis (2-D DIGE). Expression of serum proteins significantly altered between ALK inhibitor-sensitive and -resistant NSCLC was identified by liquid chromatography-tandem mass spectrometry. Differentially expressed serum proteins were confirmed by Western blot analysis. The Cancer Genome Atlas database was used to predict the protein’s expression in NSCLC. Differentially expressed proteins between ALK inhibitor-sensitive and -resistant NSCLC were identified, and markers associated with cancer progression were further analyzed in NSCLC tissue samples. Serum methylated proteins were identified by methyl-specific immunoprecipitation and confirmed by Western blot. The Cancer Genome Atlas database showed that protein abundance of cytokeratin 18 (CK18) was associated with ALK inhibitor response. In NSCLC tissue samples, CK18 expression was significantly increased in ALK inhibitor-sensitive compared with -resistant tumors. Serum CK18 levels in ALK inhibitor-sensitive NSCLC were lower than those in ALK inhibitor-resistant tumors. The difference in serum CK18 levels between ALK inhibitor-sensitive and -resistant NSCLC was further confirmed in an independent validation cohort. The experimental parameters were optimized. The system was evaluated by the analysis of tap water samples. Under the optimized conditions, the analytical performance of this method was evaluated by the recovery and RSD. Also, the analytical figures of merit were presented. The effects of interfering ions were examined by the determination of uranium(VI) in the presence of 2.5-1000 microg mL(-1) of Ca(2+), Mg(2+), Al(3+), Fe(3+), Fe(2+) and Cr(3+) at pH 8.0. The maximum enrichment factor was 64. The linear range of uranium(VI) was 0.1-1.5 microg mL(-1). The limit of detection was 0.006 microg mL(-1). The method was successfully applied to the determination of uranium in two drinking water samples and a creek water sample.
